Follistatin-likeprotein1(FSTL1)isawidely-expressed,extracellularglycoproteinthatishomologouslygroupedintotheosteonectin(BM-40/SPARC)familyofsecretedproteinsbasedonitspossessionofbothafollistatin-likeandextracellularcalcium-bindingdomain.InitiallyidentifiedasaTGF-beta-inducIBLeproteininaclonedmouseosteoblastcellline,FSTL1hassincebeenimplicatedinanarrayofcell-type-specificfunctions,suchastheregulationofproliferation,differentiation,apoptosisandmigration,aswellasanumberofBIOLOGicalprocesses,includingembryonicdevelopment,inflammatoryresponse,angiogenesis,tumOrigenesis,andimmunediseasepathogenesis.Highlyconservedacrossmammalianspeciesandwidelyexpressedinhumantissues,FSTL1canbeupregulatedthroughsignalingmediatorsoftheinnateimmunesystem,suchasTLR4agoNISTsandthearthritogeniccytokineIL-1betaviaNFκBpathways,tostimulatetheexpressionandsecretionofpro-inflammatorycytokines,includingTNF-alpha,IL-1beta,IL-6andIL-8.WhilecellsofmesenchymallineagearecapableofFSTL1production,FSTL1expressionisnotablyabsentfromcellsofhematopoieticlineageundernormalphysiologicalconditions.Macrophagesandmonocytesare,however,capableoftakingupFSTL1atsitesofinflammationwhereFSTL1stimulationcancausetheexpressionofcaspase-1anditsresultantenzymaticcleavageofactiveIL-1betafrompro-IL-1beta.WhereastheoverexpressionofFSTL1hasbeennotedasasubstantialcontributortotheprogressionofimmunediseaseslikerheumatoidarthritis(RA)andosteoarthritis(OA),diminishedFSTL1serumlevelshavebeenidentifiedasplayingasignificantpartinbothovarianandendometrialcarcinogenesis,whereitdirectlyaffectscellproliferation,migrationandinvasion.TheCHOcell-derivedrecombinanthumanFSTL1isa288-amino-acid-lengthglycoproteinwithacalculatedmolecularweightof32.7kDa;however,duetoglycosylation,proteinmigrationoccursatanapparentmolecularweightofapproximately50-55kDabySDS-PAGEanalysisunderbothreducingandnon-reducingconditions.
