Description:

Angiogenesisresearchhasfocusedonreceptorsandligandsmediatingendothelialcellproliferationandmigration.Littleisknownaboutthemolecularmechanismsthatareinvolvedinconvertingendothelialcellsfromaproliferativetoadifferentiatedstate.Microvasculardifferentiationgene1(Mdg1)hasbeenisolatedfromdifferentiatingmicrovascularendothelialcellsthathadbeenculturedincollagentypeIgels(3Dculture).InadulthumantissueMdg1isexpressedinendothelialandepithelialcells.Sequenceanalysisofthefull-lengthCDNArevealedthattheN-terminalregionoftheputativeMdg1-proteinexhibitsahighsequencesimilaritytotheJ-domainofHsp40chaperones.ItwasshownthatthisregionfunctionsasabonafideJ-domainasitcanreplacetheJ-domainofEscherichiacoliDnaJ-protein.Mdg1isalsoupregulatedinprimaryendothelialandmesangialcellswhensubjectedtovariousstressstimuli.GFP–Mdg1fusionconstructsshowedtheMdg1-proteintobelocalizedwithinthecytoplasmundercontrolconditions.StressinducesthetranslocationofMdg1intothenucleus,whereitaccumulatesinnucleoli.CostainingwithHdj1,Hdj2,Hsp70,andHsc70revealedthatMdg1colocalizeswithHsp70andHdj1incontrolandstressedHeLacells.ThesedatasuggestthatMdg1isinvolvedinthecontrolofcellcyclearresttakingplaceduringterminalcelldifferentiationandunderstressconditions.
