Description:

Originallyidentifiedasadifferentiationantigenofmesotheliomas,ovariancystadenocarcinomas,andpancreaticadenocarcinomas,mesothelinisaglycosylphosphatidylinositol(GPI)-anchored,cell-surfaceglycoproteinpredominantlysecretedbycellsofthemesothelium.Althoughmesothelinisexpressedatrestrictedlevelsbynormalmesothelialcellsofthepleural,pericardial,andperitonealmembranes,aberrantexpressionhasbeendocumentedintheaforementionedcancers,aswellasinendometrioiduterineadenocarcinomasandsquamouscellcarcinomasoftheesophagus,stomach,lung,andcervix.Proteolyticcleavageofmesothelinyieldsasoluble,polypeptidefragment-designatedmegakaryocyte-potentiatingfactor(MPF)basedonitsabilitytostimulatemegakaryocytecolony-formingactivityofmurineIL-3inmurinebonemarrowcellcultures.OriginallyisolatedfromtheHPC-Y5pancreaticcellline,MPFhasbeensuggestedtoplayaroleintheproliferationanddifferentiationofmegakaryoctyes,andtheregulationofresultantplateletproduction.WhiletheBIOLOGicalfunctionsofbothmesothelinandMPFremainspeculative,highlevelsofexpressionincanceroustissues,comparedtolimiteddistributioninnormaltissues,stronglysuggeststheirinvolvementintumOrigenesis.Bothhavebeendemonstratedtopromotetumorcellproliferation,migration,anchorage-independentgrowth,andtumorprogression,demonstratingtheirinvolvementinheterotypiccelladhesionandthemetastaticspreadofcancer.RecombinantHumanMesothelinisaglycoproteincontaining327aminoacidresidues,andhasacalculatedmolecularweightofapproximately36.4kDa.Asaresultofglycosylation,RecombinantHumanMesothelinmigrateswithanapparentmolecularmassofapproximately40-45kDabySDS-PAGEgel,underreducingandnon-reducingconditions.
