Description:

M-CSF,alsoknownasCSF-1,isafour-αhelical-bundlecytokinethatistheprimaryregulatorofmacrophagesurvival,proliferationanddifferentiation.M-CSFisalsoessentialforthesurvivalandproliferationofosteoclastProgenitors.M-CSFalsoprimesandenhancesmacrophagekillingoftumorcellsandmicroorganisms,regulatesthereleaseofcytokinesandotherinflammatorymodulatorsfrommacrophages,andstimulatespinocytosis.MCSFincreasesduringpregnancytosupportimplantationandgrowthofthedeciduaandplacenta.SourcesofMCSFincludefibroblasts,activatedmacrophages,endometrialsecretoryepithelium,bonemarrowstromalcells,andactivatedendothelialcells.TheM-CSFreceptor(c-fms)transducesitspleotropiceffectsandmediatesitsendocytosis.M-CSFmRNAsofvarioussizesoccur.FulllengthmouseMCSFtranscriptsencodea520aminoacid(aa)typeItransmembrane(TM)proteinwitha462aaextracellularregion,a21aaTMdomain,anda37aacytoplasmictailthatformsa140kDacovalentdimer.Differentialprocessingproducestwoproteolyticallycleaved,secreteddimers.OneisanNandOglycosylated86kDadimer,whiletheotherismodifiedbybothglycosylationandchondroitinsulfateproteoglycan(PG)togeneratea200kDasubunit.AlthoughPGmodifiedM-CSFcancirculate,itmaybeimmobilizedbyattachmenttotypeVcollagen.ShortertranscriptsencodeM-CSFthatlackscleavageandPGsitesandproducesanN-glycosylated68kDaTMdimerandaslowlyproduced44kDasecreteddimer.Althoughformsmayvaryinactivityandhalf-life,allcontaintheN-terminal150aaportionthatisnecessaryandsufficientforinteractionwiththeM-CSFreceptor.Thefirst229aaofmaturemouseMCSFshares87%,83%,82%,and81%aaidentitywithcorrespondingregionsofrat,dog,cow,andhumanMCSF,respectively.HumanMCSFisactiveinthemouse,butmouseM-CSFisreportedtobespecies-specific.
